Novel Astatine Methodologies for Theranostics and Radio Therapy Udbyder Vejleder Sted København og omegn Astatine is perhaps arguably the most unique element known to mankind. Discovered in 1940 by the colleagues Corson, MacKenzie, and Segrè at the University of California, it was found that no stable isotopes are available for this element, and that their natural abundancy was less than 1 gram at any given time on Earth. Despite this, for example, Astatine-211 can be produced synthetically, and has very useful properties as a radionuclide within the breath of radio therapy. Astatine-211 has a prominent electron capture decay (58%) and alpha particle decay (42%), of which can be used in radio therapy to kill cancer cells or other significant diseases. Despite these key beneficial features, the chemical and physical properties of Astatine-211 hamper research and development on this unique isotope remains scares.In this project we will develop a novel radiochemical reaction for the introduction of Astatine-211 in pharmacological relevant chemical compounds. The new methodology will allow rapid exchange of a leaving group with the Astatine-211 isotope (see figure) on a large scope of molecules and with medically relevant chemical structures. More importantly, and fundamentally crucial to astatine-compounds, the stability of these astatinated compounds will be subject to intense interrogation. Astatinated compounds are notoriously unstable due to the physical properties of this isotope, making research highly challenging but also very valuable. During the course of this project, the student will develop a novel strategy for the incorporation of Astatine-211, Iodine-125 or Iodine-131. Several different compounds with different chemical structures will be prepared to evaluate their stability in vitro and in vivo.
