AUTOSECT- Engineering yeast for insect managment Udbyder Vejleder Sted København og omegn Over millions of years, insects have evolved a finely tuned system for detecting and reacting to odorants in plants and yeast. Insect olfactory pathways sense pheromones and volatiles from plants and microbes and trigger behavioral changes in response. Since insects play critical roles as both pollinators and as destructive pests, it is of great value to increase understanding of insect olfaction.Being able to refactor the insect olfactory system in the model yeast Saccharomyces cerevisiae, AKA expressing insect olfactory receptor proteins that respond to odors in the environment in yeast, would allow us to quickly screen which odorants affect different olfactory receptors. Such a platform is expected to facilitate the design of target pesticides, bioactive foods and novel pollination systems.Ultimately, this project will use synthetic biology techniques to develop reporter assay maps by heterologously expressing insect olfactory receptor complexes in yeast to mimic insect odor-maps and create insect bioactive foods that can affect insect behaviour. This project will also afford opportunities to work with non-model strains of yeast as well as designing bioactive foods as tools? to affect the behaviour of flies and other insects.Skills that will be gained: Standard molecular cloning and microbiological techniques, yeast genetic engineering with CRISPR-Cas9, flow cytometry and FACS, droplet microfluidics using the Xdrop platform (Samplix), microscopy (epifluorescence and confocal), experimental design and data analysis.
